Preparing the kit

1. Reconstitute CJC-1295 with DAC (10mg). Add 2 mL of bacteriostatic water slowly down the inside wall of the vial and swirl gently until clear; do not shake. 10mg in 2 mL gives 5 mg/mL. The calculator has this vial preset.
2. Reconstitute Ipamorelin (10mg). Add 2 mL of bacteriostatic water slowly down the inside wall of the vial and swirl gently until clear; do not shake. 10mg in 2 mL gives 5 mg/mL. The calculator has this vial preset.
3. Draw and swab. Swab the vial stopper before each draw. Use a fresh U-100 syringe each time; the calculator converts the amount in milligrams to syringe units at the concentration prepared.
4. Store. Lyophilised vials at −20 °C until reconstitution. Reconstituted solution and pens at 2–8 °C, never frozen; reconstituted vials are used within 8–12 weeks, opened bacteriostatic water within 28 days.

Long-acting GHRH analogue with drug-affinity complex

CJC-1295 with DAC takes that same GHRH (1-29) analogue and attaches a maleimidopropionic acid group, which bonds covalently to serum albumin. The payoff is a half-life stretched from minutes to roughly six to eight days, giving a steady lift in GH and IGF-1 in place of separate pulses.

Sustained rises in GH and IGF-1 in healthy adults, after both single and repeated doses, were reported by Teichman et al. (JCEM 2006). ConjuChem's development of it for HIV lipodystrophy was halted. These days it serves as a research tool for sustained stimulation of the GH axis.

Ipamorelin

Selective ghrelin-receptor (GHS-R1a) agonist pentapeptide

Ipamorelin is a growth-hormone secretagogue of five amino acids that switches on the ghrelin receptor. What marks it out within the GHRP family is how clean it is: the first characterisation showed it releasing growth hormone while leaving the cortisol, prolactin and ACTH responses of GHRP-6 and GHRP-2 behind.

The selectivity profile was laid out by Raun et al. (Eur J Endocrinol 1998) in rats and pigs. Ipamorelin reached a completed Phase 2 trial for post-operative ileus that was not taken further. Bone density and gastric motility are the focus of the preclinical work.
